The title of the thesis was: (Study the Gene Expression of Hypoxia Inducible Factor-1 Alpha mRNA and Estrogen Receptor Index in Women and Cell lines with Breast Cancer) .
The discussion committee consisted of Prof. Dr. Mufid Jalil Awad as Chairman, and the membership of Prof. Dr. Omar Farouk Abdul Rashid, Assistant Professor Dr. Muhannad Jawad Kazim, Assistant Professor Dr. Ali Jaber Al-Mamouri, Assistant Professor Dr. Ibtisam Faris Muhammad, Professor Muayyad Imran Al-Ghazali as Member and Supervisor, and Professor Dr. Mushtaq Abdul azim witwit member and supervisor.
The student explained that the study aims to:
• First: a study of the genetic expression of the inherited HIF-1A mRNA as well as the ERI index of estrogen receptors in Iraqi women with breast cancer.
• secondly: the preparation of the nanoparticle of the medicine inhibiting the evolyms of the MTOR pathway; This polymer is a PLGA-TPGS-EV-FOL surrounded by folate molecules for the purpose of pharmacological nanoparticles bound to folate receptors with a large division in cancer cells. It has been tried on MCF-7 breast cancer cell lines to demonstrate its effect on the programmed death paths of cancer cells, through The transition of glucose sugar into the cells to feed and produce energy and the process of forming new blood vessels that supply the cancerous cells with blood to form the tumor, the study included 120 Iraqi women with breast cancer, and some of them were in the initial stage of the injury, and another section that takes chemotherapy after performing surgeries Whether or not it is done to remove the tumor from the affected breast, and another 120 other women who are healthy from this disease are close to age and the weight index has been taken as a control group .
The methods of work used to measure the levels of ERA, ERB, GLUT-1, CD44 and VEGF are the use of the ELISA technique. As for measuring the gene expression levels of the genes of the two genes HIF-1A and FOLR were done by the instantaneous quantitative PCR (qRT-PCR) after extracting the RNA The total newly drawn blood from the patient and control groups and converting it into its corresponding DNA (cDNA).
The study was divided into two parts; the first was a biochemical and genetic study, while the second was a study of the manufacture of a polymeric carrier of the drug inhibiting the mTOR pathway.
The study included the killer effect of nanomedicine compared to free non-nanomedicine on MCF-7 cancer cell lines, and it showed that there is a significant statistical difference between the percentage of cells is 3 days compared to the rate of killing 13% in the duration of one day incubation with the nanomedicine. The study concluded that preparing and modifying the nanoparticles of the polymeric nanoparticle of the medicine supplied with folate to target cancerous cells may contribute to reducing the large cost of the drug, as well as reduce the side effects caused by the free EV drug on patients, and after extensive discussion the thesis was accepted; For fulfilling the regulations for accepting university theses.
